Scalemic (S)-methylphenylphosphine oxide [(S)-2e] is shown to undergo arylation without detectable stereoerosion. The application of this method to the synthesis of novel P-chiral phosphines and PCP ligands is demonstrated.

Biomimetically inspired asymmetric total synthesis of (+)-19-dehydroxyl arisandilactone A

Complex natural products are a proven and rich source of disease-modulating drugs and of efficient tools for the study of chemical biology and drug discovery. The architectures of complex natural products are generally considered to represent significant barriers to efficient chemical synthesis. Here we describe a concise and efficient asymmetric synthesis of 19-dehydroxyl arisandilactone A – which belongs to a family of architecturally unique, highly oxygenated nortriterpenoids isolated from the medicinal plant Schisandra arisanensis. This synthesis takes place by means of a homo-Michael reaction, a tandem retro-Michael/Michael reaction, and Cu-catalysed intramolecular cyclopropanation as key steps. The proposed mechanisms for the homo-Michael and tandem retro-Michael/Michael reactions are supported by density functional theory (DFT) calculation. The developed chemistry may find application for the synthesis of its other family members of Schisandraceae nortriterpenoids.

Alpha particle-emitting radionuclides have gained considerable attention for radionuclide therapy. Astatine-211 (211At) is a promising alpha particle-emitting radionuclide. 211At is a halogen that has similar chemical properties to iodine and exhibits a half-life of 7.2 h. However, direct labeling of proteins or peptides into the tyrosine residue with 211At was shown to be impractical. Herein, we demonstrate a novel 211At-labeling method using the RGD peptide as a model peptide. An 211At-labeled RGD peptide, [211At]c[RGDf(4-At)K], was prepared from a precursor with a tributylstannyl group on the phenylalanine residue in c(RGDfK) with a radiochemical yield of 63% and a radiochemical purity of >96%, and its potential for targeted radionuclide therapy was evaluated. Based on the results of biodistribution experiments, [125I]c[RGDf(4-I)K] and [211At]c[RGDf(4-At)K] showed high accumulation in the tumor and similar biodistribution. This study provides useful information for radiotheranostics between an 211At-labeled peptide and the corresponding radioiodine-labeled peptide.

Reprogramming of mitochondrial functions sustains tumor growth and may provide therapeutic opportunities. Here, we targeted the protein folding environment in mitochondria by coupling a purine-based inhibitor of the molecular chaperone Heat Shock Protein-90 (Hsp90), PU-H71 to the mitochondrial-targeting moiety, triphenylphosphonium (TPP). Binding of PU-H71-TPP to ADP-Hsp90, Hsp90 cochaperone complex or mitochondrial Hsp90 homolog, TRAP1 involved hydrogen bonds, p-p stacking, cation-p contacts and hydrophobic interactions with the surrounding amino acids in the active site. PU-H71-TPP selectively accumulated in mitochondria of tumor cells (17-fold increase in mitochondria/cytosol ratio), whereas unmodified PU-H71 showed minimal mitochondrial localization. Treatment of tumor cells with PU-H71-TPP dissipated mitochondrial membrane potential, inhibited oxidative phosphorylation in sensitive cell types, and reduced ATP production, resulting in apoptosis and tumor cell killing. Unmodified PU-H71 had no effect. Bioinformatics analysis identified a “mitochondrial Hsp90” signature in Acute Myeloid Leukemia (AML), which correlates with worse disease outcome. Accordingly, inhibition of mitochondrial Hsp90s killed primary and cultured AML cells, with minimal effects on normal peripheral blood mononuclear cells. These data demonstrate that directing Hsp90 inhibitors with different chemical scaffolds to mitochondria is feasible and confers improved anticancer activity. A potential “addiction” to mitochondrial Hsp90s may provide a new therapeutic target in AML.

Modulation of ambipolar charge transport characteristics in side-chain polystyrenes as host materials for solution processed OLEDs

A series of side-chain polystyrenes was developed as ambipolar hosts for solution processed organic light emitting diodes (OLEDs). The series was derived from the hole-only transport host molecule 1,3-Bis(N-carbazolyl)benzene (mCP). Electron transport ability was incorporated into the host polymers by the introduction of electron-poor heterocycles (pyridine or triazine) and extending delocalization of the lowest unoccupied molecular orbital (LUMO). The materials were tested in Ir-based green OLED devices with all organic layers processed from solution. Devices with the polymer combining triazine and carbazole on its side-chain exhibited a low luminance on-set voltage of 3.0?V and a current efficacy of 28.9?cd/A, which was more than 10 times higher than for devices with the mCP-based polymer (1.6?cd/A). The increase in performance is most likely due to an improvement of charge balance in the emissive layer, showing that our ambipolar polymers are good candidates for further wet-process optoelectronic applications.

Solution processable truxene based blue emitters: Synthesis, characterization and electroluminescence studies

Truxene (10,15-dihydro-5H-diindeno[1,2-a;1?,2?-c]fluorene) derived blue emitting materials were designed and developed utilizing Suzuki and Buchwald-Hartwig type cross-coupling reactions. The target molecules T1, T2 and T3 bearing N-carbazolyl, 1-pyrenyl and N-phenyl-N-(pyren-1-yl) substituents, respectively at 2,7,12- positions, were synthesized under mild conditions by palladium-catalyzed reactions in good yields. To improve the solution processability of the material, n-hexyl chains were used as substituents at 5,5?-, 10,10?-, 15,15?- positions. From UV?vis measurements, the absorption maxima were found to be at 330 nm for T1, 356 nm for T2 and 412 nm for T3. The materials were found to be blue emitting with their emission maxima at 385 nm, 425 nm and 490 nm for T1, T2 and T3, respectively. The excited state lifetimes were investigated using time correlated single photon counting and were found to be 10 ns, 1.4 ns and 5.4 ns for T1, T2 and T3, respectively. The highest photoluminescence quantum yield was observed in the case of T2 corresponding to a value of 0.97. The compounds T2 and T3 were used as active materials for the fabrication of solution processed, single layer blue light emitting diodes, with low turn-on voltage (2?3.3 V) and Commission Internationale de l’eclairage (CIE) coordinates corresponding to (0.16, 0.23) and (0.17, 0.36) for T2 and T3, respectively.
