It is well known that beta-ketoesters are useful intermediates for the synthesis of a range of heterocyclic templates. While there are many useful synthetic methods available to access these intermediates, there are still opportunities for the discovery of useful methodologies for their construction from novel starting materials. In this regard, we report on the discovery of a facile Pd-catalyzed Mizoroki-Heck coupling of ethyl 3-ethoxyacrylate with aryl and heteroaryl halides to form substituted alkoxyacrylates which can be hydrolyzed to form novel aryl and heteroaryl beta-ketoesters.

The synthesis of unsymmetrical sulfides was carried out in high yields by the palladium-catalyzed cross-coupling reaction of 9-organothio-9-borabicyclo[3.3.1]nonane (9-RS-9-BBN) with organic electrophiles, such as iodoarenes, 1-iodo-1-alkenes, allyl carbonate and propargyl carbonate. Iodoarenes and 1-iodo-1-atkenes were smoothly converted into the corresponding sulfides at 50 C in the presence of PdC12(dppf) (3 mol%) and K3PO4 (3 equiv.) in DMF. In contrast, the cross-coupling reaction of 9-RS-9-BBN with allyl or propargyl carbonates occurred in DMF without the assistance of a base. Both reactions catalyzed by Pd(dba)2-dppf regioselectively produced allyl and allenyl sulfides in excellent yields. The scope and limitations of the reactions, as well as the effects of varying the reaction conditions, are discussed.

Comparartive Study on the Reactivity of 6-Haloimidazo[1,2-a]pyridine Derivatives towards Negishi- and Stille-Coupling Reactions

The scope of the Suzuki-cross-coupling reaction of 6-haloimidazo[1,2-a]pyridines is dependent on the availability of the (hetero)arylboronic acids. Thus, with the aim to develop expanded applications of (hetero)arylations of imidazo[1,2-a]pyridines, we investigated the Negishi- and Stille-cross-coupling reactions at the 6-position. Remarkably, attempts to aply the Negishi-cross-coupling conditions to the organozinc derivative prepared from 6-haloimidazo[1,2-a]pyridine via a lithium-zinc exchange led to the 5-phenyl compound 3 in 54 percent yield instead of the desired 6-phenyl-isomer (Scheme 1). In contrast, various commercially available halogenated five- or six-membered-ring heterocycles were efficiently coupled to the 6-(trialkylstannyl)imidazo[1,2-a]pyridine under Stille conditions (Table 2).

Metallacyclic palladium(II) complexes [Pd(L)(R3P)Cl], L = TIQDTC (1,2,3,4-tetrahydroisoquinolinedithiocarbamate), 4MpipDTC (4-methylpipradinedithiocarbamate), MPizDTC (N-methylpiperazinedithiocarbamate), R3P = Ph3P, (o-tolyl)3P, Ph2ClP, were synthesized in a 1:1 molar metal-ligand ratio. These complexes were characterized by elemental analyses, FT-IR, multinuclear (1H, 13C and 31P) NMR. The X-ray crystal structures of [Pd(TIQDTC)(Ph3P)Cl] and [Pd(TIQDTC)((o-tolyl)3P)Cl] show a slightly distorted square planar environment around the Pd(II) ion with S-Pd-S and P-Pd-Cl average bond angles of 74.51 and 92.41, respectively. These complexes were screened for cytotoxic, antifungal, anti-inflammatory and antibacterial activity. Some complexes exhibit a significant activity against fungi.
