The Pd-catalyzed S-allylation of thiols with stable allylcarbonate and allylacetate reagents offers several advantages over established reactions for the formation of thioethers. We could demonstrate that Pd/BIPHEPHOS is a catalyst system which allows the transition metal-catalyzed S-allylation of thiols with excellent n-regioselectivity. Mechanistic studies showed that this reaction is reversible under the applied reaction conditions. The excellent functional group tolerance of this transformation was demonstrated with a broad variety of thiol nucleophiles (18 examples) and allyl substrates (9 examples), and could even be applied for the late-stage diversification of cephalosporins, which might find application in the synthesis of new antibiotics. The complexes (cod)MCl2 (M = Pd, Pt; cod = cis,cis-1,5-cyclooctadiene) react with Li2(cot) (cot = cyclooctatetraene) in a 1,6-diene/diethyl ether mixture (1,6-diene = hepta-1,6-diene, diallyl ether, dvds (1,3-divinyl- 1,1,3,3-tetramethyldisiloxane)) to afford the isolated homoleptic dinuclear Pd0 and Pt0 compounds Pd2(C7H12)3 (1), Pd2(C6H10O)3·C6H10O (2′; 2: Pd2(C6H10O)3), Pd2(dvds)3 (3), and Pt2(C7H12)3 (4). When 1-4 are treated with additional 1,6-diene the equally homoleptic but mononuclear derivatives of type M(1,6-diene)2 (5-8) and with ethene the mixed alkene complexes (C2H4)M(1,6-diene) (9-12) are obtained in solution. Complexes 1-12 react with donor ligands such as phosphanes, phosphites, or (t)BuNC to give isolated complexes of types L-M(1,6-diene) (13-41), which have also been prepared by other routes. In all complexes the metal centers are TP-3 coordinated: complexes 1-4 contain chelating and bridging 1,6-diene ligands, whereas the other complexes contain a chelating 1,6-diene ligand and an eta2-alkene (5-12) or eta1-donor ligand (13-41). Of the studied 1,6-diene complexes the hepta-1,6-diene derivatives are most reactive, while the diallyl ether complexes are often more convenient to handle. The readily isolable dinuclear hepta-1,6-diene and diallyl ether complexes 1, 2′, and 4, and their mononuclear pure olefin derivatives are among the most reactive sources for naked Pd0 and Pt0. The corresponding L-M(1,6-diene) complexes are equally reactive precursor compounds for the generation of [L-M0] fragments in solution, which for M =Pd are available otherwise only with difficulty. The results are significant for the operation of naked Pd0 and L-Pd0 catalysts in homogeneous catalysis.

Incompletely condensed polyhedral oligomeric silsesquioxanes (IC-POSS) are promising building blocks for the development of organic-inorganic hybrid materials. Pd-catalyzed arylation of heptaphenyl IC-POSS has been developed to introduce various substituents on to the open moieties without using highly moisture-sensitive chlorosilane derivatives. Discovery of 6-[5-(4-fluorophenyl)-3-methyl-pyrazol-4-yl]-benzoxazin-3-one derivatives as novel selective nonsteroidal mineralocorticoid receptor antagonists

In the course of our study on selective nonsteroidal mineralocorticoid receptor (MR) antagonists, a series of novel benzoxazine derivatives possessing an azole ring as the core scaffold was designed for the purpose of attenuating the partial agonistic activity of the previously reported dihydropyrrol-2-one derivatives. Screening of alternative azole rings identified 1,3-dimethyl pyrazole 6a as a lead compound with reduced partial agonistic activity. Subsequent replacement of the 1-methyl group of the pyrazole ring with larger lipophilic side chains or polar side chains targeting Arg817 and Gln776 increased MR binding activity while maintaining the agonistic response at the lower level. Among these compounds, 6-[1-(2,2-difluoro-3-hydroxypropyl)-5-(4-fluorophenyl)-3-methyl-1H-pyrazol-4-yl]-2H-1,4-benzoxazin-3(4H)-one (37a) showed highly potent in vitro activity, high selectivity versus other steroid hormone receptors, and good pharmacokinetic profiles. Oral administration of 37a in deoxycorticosterone acetate-salt hypertensive rats showed a significant blood pressure-lowering effect with no signs of antiandrogenic effects.

Benzoyl Fluorides as Fluorination Reagents: Reconstruction of Acyl Fluorides via Reversible Acyl C-F Bond Cleavage/Formation in Palladium Catalysis

This report describes the formation of value-added acyl fluorides by means of palladium-catalyzed acyl-exchange reactions between acyl fluorides and acid anhydrides. This method allows using a simple and commercially available acyl fluoride, benzoyl fluoride, as the fluoride source for the easy and efficient preparation of a variety of more complex acyl fluorides. The results of this study suggest that this reaction proceeds via a reversible acyl C-F bond cleavage/formation at the palladium center.

Palladium-Catalyzed Domino Heck/Silylation Reaction for the Synthesis of (2-Oxoindolin-3-yl)methylsilanes via Trapping of the sigma-Alkylpalladium Intermediates with Disilanes

A palladium-catalyzed domino Heck/silylation reaction of acrylamides with hexamethyldisilane is presented. This transformation, which involves the formation of a sigma-alkylpalladium intermediate, allows direct access to functionalized (2-oxoindolin-3-yl)methylsilanes in good to excellent yields. Furthermore, this strategy could be utilized for the synthesis of (2-oxoindolin-3-yl)methylstannanes from hexamethyldistannane. (Figure presented.).
