Polythiophenes (PTs) featuring di(ethylene glycol)-substituted 2,5-thienyl diketopyrrolopyrrole (DG-TDPP) moieties as conjugated units in the polymer backbone and tert-butyl-substituted triphenylamine (tTPA)-containing moieties as pendant units have been synthesized through Stille coupling. Incorporating the electron-deficient DG-TDPP moieties within the polymer backbone and appending the tTPA units promoted charge balance and efficient conjugation within the extended conjugated frameworks of the polymers, resulting in lower band gap energies and red-shifting the maximum UV-Vis absorption wavelength. The influence of the DG-TDPP content on the optical, electrochemical, and photovoltaic (PV) properties of the polymers has been studied. Incorporating a suitable content of DG-TDPP moieties in the polymer backbone enhanced the solar absorption ability and conjugation length of the PTs. The PV properties of bulk-heterojunction solar cells based on PT/fullerene derivatives improved after incorporating DG-TDPP units in the backbones of the side chain-conjugated PTs.

As part of an ongoing effort at Amgen to develop a disease-modifying therapy for Alzheimer’s disease, we have previously used the aminooxazoline xanthene (AOX) scaffold to generate potent and orally efficacious BACE1 inhibitors. While AOX-BACE1 inhibitors demonstrated acceptable cardiovascular safety margins, a retinal pathological finding in rat toxicological studies demanded further investigation. It has been widely postulated that such retinal toxicity might be related to off-target inhibition of Cathepsin D (CatD), a closely related aspartyl protease. We report the development of AOX-BACE1 inhibitors with improved selectivity against CatD by following a structure- and property-based approach. Our efforts culminated in the discovery of a picolinamide-substituted 3-aza-AOX-BACE1 inhibitor absent of retinal effects in an early screening rat toxicology study.

In order to investigate the relationships between chemical structures of conjugated polymers and their properties with respect to photovoltaic applications, two new copolymers consisting of an electron-deficient tetrazine unit and either an electron-rich carbazole (PCTz) or fluorene (PFTz) along with thiophene spacers were synthesized. We report on the synthesis, spectroscopic and scintillation properties of three new pyrazoline core based fluorophores. Fluorescence properties of the fluorophores have been studied both in a solution state and in a solid polyvinyltoluene (PVT) resin matrix of different porosity. The synthesized fluorophores were found to be promising candidates for application in plastic scintillators for detection of ionizing radiation (alpha, beta particles, gamma rays and neutrons) and demonstrated superior efficiency in comparison to the existing commercially used fluorophores (2-(1-naphthyl)-5-phenyloxazole (alphaNPO), 9,10-diphenylanthracene, etc.). Moreover, the suggested synthetic route allows functionalization of the fluorophores with a vinyl group for further covalent bound to the PVT or other vinyl polymer matrices, which dramatically improves chemical stability of the system simultaneously improving the photoluminescence quantum yield. Possible mechanisms of the enhanced scintillation properties are discussed based on preliminary quantum mechanical calculations and spectroscopic characteristics of the fluorophores under study.

We synthesized solution-processable iridium complexes having bulky carbazole dendrons, fac-tris[2-{3-(3,5-bis(3,6-di-n-butylcarbazol-9-yl)phenyl)Phenyl)pyridine]iridium (III) (mCP)3Ir and fac-bis[2-{3-(3,5-bis(3,6-di-n-butylcarbazol-9-yl)phenyl)phenyl}pyridine][2-{3-(3,5-di(4-pyridyl)phenyl)phenyl}pyridine]iridium (III) (mCP)2(bpp)Ir. Photoluminescence quantum efficiencies (PLQEs) of (mCP)3Ir and (mCP)2(bpp)Ir in their diluted solutions were 91% and 84%, respectively. They showed high PLQEs of 49% for (mCP)3Ir and 29% for (mCP)2(bpp)Ir even in a neat film. The triplet exciton energy level of the dendronized ligand (2.8 eV), 2-[3-{3,5-bis(3,6-di-n-butylcarbazol-9-yl)phenyl}]pyridine 10, and the dendron (2.9 eV), 3,5-bis(3,6-di-n-butylcarbazol-9-yl)benzene 7, are enough higher than that of the core complex Ir(ppy)3 (2.6 eV). External quantum efficiency (EQE) of single layer light-emitting device with (mCP)2(bpp)Ir was much higher than that of (mCP)3Ir because of better affinity of (mCP)2(bpp)Ir to cathode metal. When an electron transporting and hole-blocking material was used, the EQEs of double layer devices were dramatically improved to 8.3% for (mCP)3Ir and 5.4% for (mCP)2(bpp)Ir at 100 cd/m2.
