PET study using [11C]FTIMD with ultra-high specific activity to evaluate I2-imidazoline receptors binding in rat brains

Introduction: We recently developed a selective 11C-labeled I2-imidazoline receptor (I2R) ligand, 2-(3-fluoro-4-[11C]tolyl)-4,5-dihydro-1H-imidazole ([11C]FTIMD). [11C]FTIMD showed specific binding to I2Rs in rat brains having a high density of I2R, as well as to I2Rs those in monkey brains, as illustrated by positron emission tomography (PET) and autoradiography. However, [11C]FTIMD also showed moderate non-specific binding in rat brains. In order to increase the specificity for I2R in rat brains, we synthesized [11C]FTIMD with ultra-high specific activity and evaluated its binding. Methods: [11C]FTIMD with ultra-high specific activity was prepared by a palladium-promoted cross-coupling reaction of the tributylstannyl precursor and [11C]methyl iodide, which was produced by iodination of [11C]methane using the single-pass method. Dynamic PET scans were conducted in rats, and the kinetic parameters were estimated. Results: [11C]FTIMD with ultra-high specific activity was successfully synthesized with an appropriate level of radioactivity and ultra-high specific activity (4470±1660 GBq/mumol at end of synthesis, n=11) for injection. In the PET study, distribution volume (VT) values in all the brain regions investigated whether I2R expression was greatly reduced in BU224-pretreatead rats compared with control rats (29-45% decrease). Differences in VT values between control and BU224-pretreated rats using [11C]FTIMD with ultra-high specific activity were greater than those using [11C]FTIMD with normal specific activity (17-34% decrease) in all brain regions investigated. Conclusion: Quantitative PET using [11C]FTIMD with ultra-high specific activity can contribute to the detection of small changes in I2R expression in the brain.

Palladium-Catalysed Decarboxylative Generation and Regiodivergent Prenylation of 2-Azaallyl Anions

Reaction conditions for the palladium-catalysed decarboxylative generation and regiodivergent prenylation of 2-azaallyl anions to form either the branched or linear prenylated benzylic imines from prenyl 2,2-diphenyl-glycinate imines are reported. For certain substrates, formation of side products arising from a competitive elimination/protonation pathway could be reduced by the addition of various basic additives. The optimal basic additive and reaction concentration were found to be highly substrate dependent. Geranyl esters also proved to be viable substrates for the corresponding regiodivergent decarboxylative geranylation reactions. Preliminary efforts toward an enantioselective variant are also presented. (Figure presented.).

Synthesis and Characterization of Isoindigo-Based Polymers with Thermocleavable Side Chains

Stability of bulk heterojunction films for organic solar cells is a critical factor for commercial viability. One method to stabilize these films is to include cleavable side chains, which reduce the solubility of the polymers when removed. In order to study the stabilizing effect of cleavable side chains, a series of random copolymers using isoindigo with 0, 10, 20, 50, and 100% thermally cleavable side chains based on the tert-butyloxycarbonyl (t-BOC) group are synthesized. The polymers show a distinct one-step thermal cleavage of the side chains, with no separable dealkylation and decarboxylation steps. The thermal stability in film is studied with transmission electron microscopy and atomic force microscopy. The polymer with all t-BOC side chains on isoindigo significantly improves thermal stability with regard to crystal growth and phase separation in film. These results suggest BOC-substitution can be used for large scale processing to produce insoluble polymer films with a high degree of thermal stability.

Biphenyl Sulfonic and Disulfonic Acids with Perfluorinated Alkyl Residues

Sulfonic acids serve as interesting, yet not intensively studied alternatives to carboxylic acids as linker units in coordination polymers. In this study, we present the synthesis of hybrid sulfonic acids with polar, rigid biphenyl moieties and disordered highly fluorinated alkyl chains. Consequently, eight biphenyl sulfonic and disulfonic acids with one or two perfluoroalkyl chains were prepared. The key steps within the synthesis include two palladium catalyzed C?C bond formations: The perfluoroalkyl residue is installed by Heck reaction (up to 91 %) using 1H,1H,2H-perfluorinated alkenes (C6 and C8) and the biphenyl unit is established by a Suzuki coupling (up to 88 %) of boronic acids bearing one or two protected thiol groups. Finally, these thiol groups are converted into the sulfonic acids by N-chlorosuccinimide mediated oxidation followed by hydrolysis of the respective sulfonyl chloride.

The stilbene scaffold is a basic element for a number of biologically active natural and synthetic compounds, and it is considered as a privileged structure. Stilbenes exemplified by resveratrol, combretastatin A-4 and pterostilbene are of significant interest for drug research and development because of their potential in therapeutic and preventive application. Resveratrol, present in grapes and other food products, plays a role in the prevention of several human pathological processes and has been suggested as an anticancer agent. Moreover, recent evidence has revealed its potential effect on the aging process, diabetes and neurological dysfunction. Combretastatin A-4, from the bark of South African bush willow Combretum caffrum, also shows significant antitumor activity. Pterostilbene is closely related to resveratrol, sharing the same unique therapeutic potential as anti-inflammatory, antineoplastic and antioxidant agent. Therefore, research and development of stilbene-based medicinal chemistry have become rapidly evolving and increasingly active topics covering almost the whole range of therapeutic fields. In the present review, we provide an overview of the role of stilbenes in medicinal chemistry. In this context, we highlight the chemical methodologies adopted for the synthesis of stilbene derivatives, and outline the successful design of novel stilbene based hybrids in the field of cancer, Alzheimer’s and other relevant diseases. This information may be useful in further design of stilbene-based molecules as new leads for the development of novel agents with clinical potential or as effective chemical probes to dissect biological processes.
