Facile Synthesis and Self-Assembly of Zinc (2-Diethoxyphosphorylethynyl)porphyrins

The synthesis of zinc (2-diethoxyphosphorylethynyl)porphyrins and their self-organization in solid state and in solution are reported. Copper-cocatalyzed Sonogashira reaction involving bromoporphyrins and diethyl ethynylphosphonate was used to prepare meso-substituted derivatives Zn4 and Zn5 bearing one or two ethynylphosphonate substituents at the macrocycle. In contrast, copper-free Sonogashira coupling of 2-bromoporphyrin with diethyl ethynylphosphonate is more convenient to prepare porphyrin Zn6 bearing this substituent at the beta-pyrrolic position of the macrocycle. The structures of complexes Zn4a¡¤C7H8, Zn4a¡¤CHCl3, Zn4c and Zn5b¡¤CH2Cl2 in crystals were elucidated by X-ray diffraction analysis. The supramolecular self-assembly through weak coordinative binding of the phosphoryl group of one porphyrin molecule to the zinc(II) ion located in the center of another macrocyclic cavity is observed in all crystals. However, ZnII-porphyrinates Zn4a and Zn5b exist in the solid state as mutually coordinated dimers while complex Zn4c displays a 1D polymer structure. In all dimer complexes, neighboring porphyrin rings are spatially separated, with Zn?O and P=O bond lengths being similar, while the interplanar porphyrin distance is variable, depending on weak intermolecular interactions in the crystals. The self-organization of (phosphorylethynyl)porphyrins is also observed in weakly polar chloroform and detectable by NMR and UV/Vis spectroscopy. The mutually coordinated dimer of electron-deficient porphyrin Zn5b was observed in 5 ¡Á 10?3 m chloroform solution even at room temperature. The kinetic lability of this supramolecular complex was proven by NMR spectroscopy. The associates of more electron-donating porphyrins Zn4a and Zn4c are less stable, and self-assembled dimers become the predominant species in 5 ¡Á 10?3 m chloroform solution only below 225 K. The self-assembly of porphyrin Zn4a and Zn5b in more diluted solutions (up to 10?6 m) was also investigated by UV/Vis spectroscopy.

Complete details of a convergent asymmetric synthesis of oleandolide (1), the aglycon of the macrolide antibiotic oleandomycin, is described. The synthesis has been achieved through the assembly and coupling of the left- and right-hand subunits 12 and 38, respectively. These subunits were prepared from chiral silane-based asymmetric crotylation reactions to control the stereochemical relationships. The left- and right-hand subunits (C1-C7 and C8-C14) were brought together through a Pd(0)-catalyzed sp3-sp2 cross-coupling reaction between the zinc intermediate 40 and vinyl triflate 38 to give 27. This product was converted to seco acid 42a and cyclized to lactone 35 under Yamaguchi conditions. This material was then epoxidized with m-chloroperbenzoic acid (m-CPBA) to install the correct C8 epoxide as a single diastereomer, which after a short deprotection sequence completed the synthesis of oleandolide.

All of the complexes display moderately strong luminescence at room temperature with lifetimes in the range of 10-55 ns. The effect of solvents on the absorption and emission spectral behavior of the complexes has also been studied in detail. The anion sensing properties of all the complexes have been studied in solution using absorption, emission, and 1H NMR spectral studies and by cyclic voltammetric (CV) measurements. It has been observed that the complexes 1, 3, and 4 act as sensors for F-only, whereas 2 acts as sensor for F-, AcO-, and to some extent for H2PO4-. It is evident that in the presence of excess of anions deprotonation of the imidazole N-H fragment(s) occurs in all cases, an event which is signaled by the development of vivid colors visible with the naked eye. The receptor-anion binding/equilibrium constants have been evaluated.

The synthesis and ligand-centered redox chemistry of palladium complexes bearing two potentially bidentate verdazyl ligands is explored. Reaction of 1,5-diisopropyl-3-pyridin-2-yl-6-oxoverdazyl radical 1 with Pd(NCMe)4¡¤2BF4 gives a complex containing two coordinated verdazyl radicals. The structure of this complex consists of one verdazyl bound to Pd in a bidentate mode and the second verdazyl bound in a monodentate fashion through the pyridine substituent; the fourth coordination site is occupied by a solvent molecule (acetonitrile (3) or dimethyl sulfoxide (4)). Two-electron reduction of this complex with decamethylferrocene affords a bis(verdazyl) palladium complex (5) in which both verdazyls have been reduced to their anionic state and are both bound to Pd in bidentate manner. Complex 5 can be independently synthesized by a redox reaction between 1 and Pd2(dba)3. Reduced complex 5 can be re-oxidized to 3 or 4 with AgBF4; in contrast, oxidation with PhICl2 leads to ligand dissociation, ultimately giving radical 1 and a mono(verdazyl)dichloropalladium complex 2. One-electron oxidation using PhICl2 produces a formally “mixed valent” (in ligand) bis(verdazyl)chloropalladium complex (6) with one bidentate verdazyl anion ligand and one monodentate (pyridine-bound) verdazyl radical. Attempted protonation of the verdazyl ligands in complex 5 leads to complete ligand dissociation and protonation of both the tetrazine and pyridine moieties; deprotonation regenerates 5. Subsequent air oxidation of the tetrazane/pyridinium cation (formed as a tetrachloropalladate salt) leads to re-coordination of the verdazyl ligands to give 6 initially, but ultimately produces a combination of free radical 1 and 2.
